PE-22-28 is a synthetic peptide derived from the natural propeptide of sortilin. Acts as a selective inhibitor of the TREK-1 potassium channel.
Mechanism — TREK-1 antagonism increases serotonergic activity in the dorsal raphe nucleus. Produces antidepressant effects in animal models with onset of days rather than the weeks required for SSRIs.
Use case — Investigational for treatment-resistant depression. No approved human use. Research dosing 200-500 mcg subcutaneous.
Caveats — Human safety and efficacy unknown. Don't combine with serotonergic drugs without medical oversight. Wait for proper trials.
